a reviewer's comment on my manuscription show that wound healing assay and matrigel invasion assay could not prove cancer cell metastatic potential change.i donn't know the reason, could anyone help me? thanks a lot.
The metastatic potential is a very complex biological interplay in which amazing interactions occur between metastasizing cancer cells, their cancer-associated cells and the pressure of the tumor microenvironment (see the attached articles).
Some isolated cancer cell growing alone (without cancer-associated cells (fibroblasts, cells from the immune system, cells from the vascular (blood AND lymphatic) system, etc., etc.), in a 2D environment, without hypoxic pressure, and prisoners of plastic stuffs cannot reconcile / recapitulate the "actual" metastaic behavior. This is the reason why it is so easy to kill "metastatic cancer cells" in plastic flasks, dishes, etc... but so difficult in the clinical setting.
If you want to study "in vitro metastatic effects" you must AT LEAST use 3D models, some examples of which are provided here attached.
@Robert, thank you very much for this explanation. your words open my mind, thanks again! tumor microenviroment is truely a big problem should to be focoused on. it raises my interest.
