I am experimenting on the effect of salinity in Arabidopsis. I had worked with the same brand of MS medium before, and the seedlings had grown green as usual. But then I used another package of medium (new and stored in a laboratory fridge) and the seedlings turned white in about a week. I do not know which could be the cause of this problem.
Is it from the same batch of seeds? Since you said the seeds on the negative control (without salt) turned white, I guess it might because of the seeds (if the culture conditions are fine) instead of culture medium.
Yes, I used the same batch of seeds than when they grew green. I am not the only one in the lab observing this in their seedlings.
Are you using a Arabidopsis wild type or the seeds have been modified by tansgenesis or mutation or some other kind of genetic modifications? Have you checked the expiring date of culture medium instead it's have been storaged under appropiated conditions? Is it possible that some environmental conditions (mainly light regime) have changed regarding when you obtained green plants?
1. Re-prepare another batch of MS medium, and try it again. Make sure all components used are correct.
2. Make sure the 'sterilization' procedure for the seeds is correct. (too harsh, ex. high concentration or prolonged washing in alcohol or bleach can hurt the seeds).
Thank you all for your suggestions, I'm repeating the experiment with a batch of newly harvested seeds and with a less harsh sterilization procedure.
