Hello, I am having trouble achieving clear plaques when performing double-layer agar technique. Can any one explain how to obtain distinct plaques.?

The problem I am facing that the plaques are not as clear or well defined as well defined as I would like them to be.They appear smudged or blurry making it challenging to accurately quantify and analyse the results

It might just be the phage that you are using, some phage give really clear plaques and other phages the plaques are fuzzy and turbid. The other possibility has to do with your plates. Be sure your plates are fresh and not too dried out. Also be sure you are not using too much top agar for the top layer, or too many cells or cells that are too old.
