Use of antioxidant mixture (ascorbic acid 0.1 g/l + citric acid 0.15 g/l) to reduce browning of plant tissue from phenolics during explants superficial disinfection and use ascorbic acid and/or activated charcoal on the initiation culture media. Also the first week putting in darkness and progressive increase the light. Frequently change the culture medium
In tissue culture the common practice is to add activated charcoal (~ 1 g/l) which helps absorbing the secreted phenols. Antioxidants can also be added to prevent oxidation of phenols (which is the real problem for plant) as Ram Krishan suggested. Another point is to avoid inducing plant secreting of phenols, including reducing the light (activated charcoal also helps), lowering temperature, and limiting injury to plant tissue by making clean cut during subculture.
