I am looking into remove organic compounds from water by adsorption using porous materials. My question is on how to calculate the required amount of adsorbent and selecting of concentration of adsorbate? And also, after adsorption, the adsorbed amount will be measured by using HPLC. When the calibration is drawn, the standards should be also treated with conditions used such as temperature and duration of shaker in adsorption?

Depend on your organic target pollutants in real wastewater design your experiments. For example in real wastewater pharmaceuticals are present at low concentrations (10-100 ppm). So it is better to choose the concentration depend on real wastewater situation.
Regarding adsorbent it is difficult to predict the quantity of the dose required because sorption performance depends on the kind of adsorbent you are using (activated carbon, chemically modified, hybrid adsorbent). Check the previous literature about the adsorbent that you are using, which will be useful to determine appropriate quantity of adsorbent.
