The HE stain we use is very unstable. When a slide is stained in the morning and the same slide (at a deeper cut) is stained in the afternoon there is a major difference in the color. The one from the afternoon is much lighter than the one from the morning. Please help?

Hi,
You would better stain multiple slides at a time, for example slides of same experiment with different treatment groups. Secondly, try to use fresh staining solutions and buffers for every experiment. Also, strictly maintain incubation time in each solution. Hope you will overcome the current problem.
Hasan Al Faruque We do stain multiple slides at a time on the Sakura tissue Tek prisma plus, so that is a max of 75 slides at a time. But in the afternoon there was a request for a deeper cut of the slide form that morning. So we have an automated system and a strict incubation time of an hour. But still the slides that are stained later on the day are lighter than in the morning. Its a problem that appears often.
