My thesis is about isolation of biosurfactant producing halophilic streptomyces, and now I have isolated some bacteria that look like streptomyces (chalky white colony ). I am trying to find out if they produce biosurfactants or not, so I added oil to starch casein medium as a carbon source, then used its supernatant for an oil spreading test. Will it work ?
Hi Farzaneh, I think it could work and you just try it. If oil interferes growth of your Streptomyces, you can exclude oil in the medium. Then, test oil spreading activity of the culture supernatant.
adding oil to the medium might work. If not you can screen the culture broth for its surface tension, oil spreading, emulsification of oil and drop collapse activity. If you have any problem in growing Streptomyces in oil (crude oil) containing medium, you could try some light oils, example, vegetable oil and screen the culture broth for surface tension, oil spreading, emulsification of oil. The surface tension measurement is a direct proof for biosurfactant production.
