Hi I have tried the uptake control and it does not work on NK cells but it does on T cells but only a 30% of the cells take it. We used both frozen and freshly extracted PBMCs, there is no difference.
We diluted the uptake to 1/10 and used 4ul with a concentration of 120 000 cells/ 200ul RPMIc
In our hands, there is uptake, but the localisation is endosomal, leading to degradation which leads to a fluorescent increase which does not correlate with mRNA level.
