I've read a few papers which have used the chromatic shift of congo red to quantify beta-1,3 and beta-1,3-1,6 glucans from mushrooms, however I can't seem to get this to work. Has anyone else tried this and been successful? If so, could I have some details of the protocol?

Yes sir, we can quantify beta glucan with congo red. Congo red has been optionally sensitive towards beta glucan. For more information, you can check out this paper
