I have not done it on cell lysate before, but I noticed that different tissue samples gave different catalase activities (e.g. I need to dilute heart tissue more than brain tissue, and both require more dilutions than plasma samples). I assume the same applies for different cell lines that you use in your in vitro experiments.
I would suggest take some aliquots of the reagents from the kit and test them on your additionally harvested cell lysate. Find the optimised protein concentrations for your type of cells and use that for your subsequent experiments. The well plate that comes with the kit is not coated so you can use any 96-well plate for your tests and go back to the kit once you are ready. Good luck!
