We are trying to characterize neutrophil sub-types from the blood and BAL fluid of human patients and noticed that some patients have unusual neutrophil populations (CD16+HLA-CD14-) that appear as two separate populations on FSc SSc. Any idea what these separate populations may be (we have ruled out doublets)?

We did some back-gating using Fsc height and width for macrophage-differentiation from doublets/debris/aggregates. That worked fine, we were able to rule out the debris. We found CD14-CD16+(+) cells being a separate population in our cd14cd16 analysis. We did not perform HLA analysis. I'm not sure because your cells (being HLA-) and I am not totally familiar with neutrophil characteristics, if the cells you are looking at could be NK-cells.
95% of CD16+ cells in human blood are neutropgils as they are double positive for CD16 and CD66b. However the remaining CD16+ (5%) cells could be a monocyte subset. Make sure the other population is not monocytes. I would suggest double stain your cells with CD66b and CD16 and then consider double positive as neutrophils.

We have found distinct populations of circulating neutrophils having different FSC. The data suppose to be published soon...
