Has anyone tried DNA capture hybridisation protocol on Covaris ultrasonicator? I am trying to cut down hybridisation time from 16 hours to jsut few and looking into using sonication to speed it up with kinetic energy. Anyone tried that yet?
Wouldn’t you risk damaging your library by sonicating at that stage? What settings were you planning to use? I wouldn’t recommend this.
There are some companies like Twist that offer a quick 4h capture if you’re interested in cutting down the time, but I haven’t looked at the efficiency compared to the standard 16h.
In this case, it won't be shearing, but increasing the reaction kinetics, making molecules move quicker and probes binding to the prepared libraries better. I know Twist and IDT and other providers offer 4hr hybridization times, but that is still hald of a working day, and then the washes after make it still too long if you want to qc and load pool as well in real life lab with multiple set ups a week.
