I have problem with the mCP membrane filtration method for Clostridium perfringens. The issue is that when I expose the filter membrane with presumptive Clostridium colonies to ammonium hydroxide, everything turns pink and I test the media itself, which also changes color according to the pH change. I asked the chemistry people, and have been told that the phenolphthalein diphosphate should also be able to indicate colors due to pH change in the same way as free phenolphthalein. Therefore, I do not know how can we confirm the Clostridium colonies from observing the pink color showup? Because it is likely that the pink color is from the phenolphthalein diphosphate itself in the media not from the free phenolphthalein that is cleved by phosphatase produced from Clostridium perfringens.
