I have brain tissues that are fixated for a few years and I plan on staining them using fluorescent Neu-N antibody. I know that it usually does not work without antigen retrieval. I could not find a reliable protocol for that. What do you think is the best way of working with those tissues? A detailed protocol would be much appreciated.

Histological and immunohistochemical techniques yielded repro- ducible staining results when applied to human brain tissue stored in formalin for long periods; so they can be used in well preserved biobank material which are the most targeting research areas in neuropathology.

Automated machine will not be a issue. as they have used 20 year old samples it may we good to get thinner slices as far as possible. Locally if any institute has one can request also. but you should get results even if slices are thicker. with best wishes.
