I have recently been working with Ga-68 labeled biomolecules and I have only 50% recovery from my borosilicate glass vial after the product has been dried at 50 degree C under a stream of nitrogen gas (to evaporate acidified ethanol). I am trying to dissolve the final dried product in 1X PBS for formulation, but facing huge losses.
Is there a pre-treatment of the vial that I can do or anything that I can add to the product to assist a greater recovery, and would still be safe to inject into small animals?
Yes, I have experienced a similar problem with a Tc-99m-labeled peptide. Pre-treating the tube with water or ethanol, or addition of some cold mass (unlabeled peptide) in the solution didn't work. The only think that worked was not completely drying down the solution-I was drying it until one drop (50 μL) and I had much better recovery after diluting with PBS (1-2 mL).
However, in your case there will be some residual ethanol left (and HCl?). In my opinion it is not a big deal since a small amount of ethanol (
Thank you for your answer, George. That is precisely what I tried and it seems to work. I did not take my product down to the complete dryness and the recovery in PBS was greater than 90%. I also pre-blocked the borosilicate glass vial with a 1% BSA solution for 10 minutes, prior to elution and dry down, that seemed to help too.
I am glad it worked. Sticky radiolabeled compounds are often a problem
