I was wondering if someone has experience in unpacking an UHPLC column (2.1mmx150mm) to collect the beads from inside.
A part from removing the frit end of the column, is it necessary to use a LC pump to pressure out the beads?

Hi, this is not an answer. Just wondering why would you want to crack open a packed column and extract its packing material?
I have an experience to open column without cap, wherein fortunately, there was no problem in qualty. But i did not use it for a chemical analysis

After you open the column, did the beads inside fall down easily? or did you need to apply pressure with a LC pump?
- Yes, with the column outlet end-fitting removed, you use the HPLC pump at a low flow rate to gently extrude out the silica support material (as a slurry) into a clean beaker. Rinse with solvent, then centrifuge to collect. --This is done routinely when packing HPLC columns for use. I have done so many times. For Example: When packing new HPLC columns, sometimes the QC test results are disappointing. When this happens, we unpack the column, make up a new slurry, then re-pack the support again, flush and test the column (until we achieve the results needed).
BTW: Without the proper HPLC column packing equipment, you will never be able to re-pack an HPLC column correctly again, but I imagine from your question that you have no intention of doing that.
