Hello, I was wondering if anyone has a well established growth media recipe for Halobacterium salinarum. I mainly work with these species and Halorubrum as well. In the lab, we grow them in the 4M NaCl media with yeast extract, but we have some concerns regarding debris/flakes coming up in the media.

Our growth media contains NaCl, MgCl2, KCl, MgSO4, sodium citrate, CaCl2, glycerol, and pH is calibrated using NaOH. I've tried the autoclaving and filtering it through .02um, however, as some time passes, flakes or debris appear no matter If the media contains cells or not. I was wondering if the flakes start forming through the salt interactions or something else, but it certainly interferes with measuring the population density.

We've also tried interchanging some components, but it doesn't seem to be helping.

Can anyone help with this?
