After giemsa staining of cultured leukocyte arrested at metaphase, I found this under light microscope. I doubt that these are chromosomes. What are these? Did the experiment fail?
Despite the quality of photo is wrong, I think there are not chromosomes but fund grime only. As Dr. Mordadi said, the chromosomes with Giemsa stain are red. In addition, normally in the microscopic field not only metaphases should be present but even ruined lymphocytes (like Gümprecht shadows). I suggest to you a preventive control (before staining) of culture by observing this microscopically (1 drop under cover slip x40 magnification phase contrast)
