Hi everyone.

I am trying to create GUVs with the electroformation method. In my same lab this was done but several years ago and I have not much details on the protocol. Anyway I found a number of them on the internet and tried. I am using tin oxide slides to see through during formation and have an electrical contact.

First time, nothing. Second time, I do see spherical shapes (about 100 microm in size) close to the upper Tin oxide slide and towards the edge of the chamber. However, at the confocal microscope it is not clear to me that these are not air bubbles. They formed quite soon during the voltage ramp.

Have you suggestions on the tricks essential to get the GUV formed with this method? I mean the critical details (like fresh lipids - not stored for months in the freezer - , etc..).
