Hi Everyone

I have been running QPCR for my tissue samples. On each plate I run a control which has NONO with cDNA made from cells. When I ran NONO for all my samples (+ control), the Cq values are comparable. However I recently ran GAPDH as another housekeeping gene to confirm my results (I am analyzing via the Delta Delta Cq method), cross the 4 plates that I have run, the GAPDH for the control cell cDNA is about 2-3 Cq lower (~18-19ish) vs the Cq for the tissue is ~20-21. Is it possible that GAPDH is expressed differently in cells and tissues and this would account for the difference in Cq values? Any input is appreciated. Thanks
