We are trying to increase the abiotic stress tolerance in rice by introducing certain stress tolerant genes via Agrobacterium-mediated transformation. The question I have is related to our plasmid construct. It has two selective markers, a kanamycin resistance gene (Npt1) and a glufosinate resistant gene (bar) as bacterial and plant selective markers, respectively. A CAMV35s has been used as the promoter for bar gene. Can 35s promoter function in Agrobacterium as well?
The answer is no, CaMV 35S promoter will now work in agrobacterium. Anything outside Left Border (LB) and right border (RB) of a binary vector will not be transcribed and thus will be un-translated in Agrobacterium. Because the promoter of prokaryotes and eukaryotes have evolved differentially and transfer of eukaryotic promoter into prokaryotes will not cause an efficient gene expression. However, there are some reports that established the CaMV 35S promoter is not only active in plants but also in the gut bacterium E.coli.
