I have a technical question regarding the measure of the SSC.

Following some treatment, my cells are becoming bigger (I can see it with the microscope) and by FACS the FSC increase, compared to Untreated cells. Our hypothesis is that this treatment increase the expression of all proteins in the cells, along with increasing actin polymerisation.

However, looking at the SSC, this one is decreased compared to Untreated cells.

My question is:

—> lets imagine a cell with a internal complexity (granularity) of 5 (lets say it contain 5 "granules")

—> Then following the treatment, the cytoplasme of this cell increase but not the number of granule. Overall, the size increasing, the FSC increase too. But what about the SSC: is it still =5 (since there is the same number of granules, i.e. 5) or is it decreasing because the "density" of the granules decreased ?

Says otherwise, could the diminution of SSC I am observing be a consequence of a "dilution" of the granules or not? If not, then it means that following my treatment my cells are losing granules. If yes, SSC decrease is likely to be only a consequence of cytoplasmic increase.
