I am extracting the fungal endophytes secondary metabolites. I am following PD broth method for the separation of mycelial mat and culture filtrate extracts hence according to literature i am following this method but in some literature they have followed continuous shaking but some followed static condition and kept in darkness under 23'C for 9 to 10 days.
Can anybody tell me why should we keep under darkness and why only for 9 to 10 days why not we keep it for 15 to 20 days. i know that by keeping in darkness stress will create by that spore production will happens fast , but what about the time period.

Metabolites are usually produced in the late log and lag phase, and after some times it also starts degrading. Therefore, time of harvesting the metabolites is crucial to get the maximum yield. Also this time will differ for different fungi and therefore, you should first standardize the kinetics of the metabolite production by the fungi of your interest and then decide the time for incubation and metabolite harvest.
