While doing PCR if someone finds perfect band for specific gene by using DNA template which was Extracted by boiling method, would the Extracted DNA template be good for sequencing? Or should the person use the DNA template which was extracted by Extraction Kit?
It will always provide a better sequencing results when you use genomic kits for DNA extraction! Better still, purify your DNA products to obtain finer chromatogram.
Kits were developed for easy use of methodologies and therefore are all dependent on laid down protocols in research materials and/or texts. It involves getting all the required materials for a particular protocol into one space. By so doing the chances of not finding a particular component of a protocol are slim. The not-too-brilliant operators could also use it for commercial purposes. However, if you have all your materials available and are skilled enough to attempt the protocol with ample time on your hands, coupled with the necessary research perseverance, I don't see why not.
DNA extraction y boiling method is sufficient for routine PCR detection, however, if you intend to conduct other downstream evaluation, it is essential to use a hugely purified template DNA, preferably extracted using kits which usually remove all remnants of unamplified segments, primers and other impurities like proteins
