WHEAT:
Take about 15-20 cores by a soil sampler of about 1" diameter to 6" depth, within about 2 meter diameter of a selected location. Mix all cores. This forms a composite. And from this mix composite, we take out about 100 gr to send to lab.
Sounds OK?
Now some questions:
1. Can we use that sample for pH also?
2. I believe EC has no meaning or use?
3. Are there more than one major varieties of wheat in the Multan region? Do they have Same root zone depth or different depths? or we should modify the depth according to the variety of wheat?
4. Should we take additional cores from 6" - 12" also for wheat? any reason?
Another issue would be fertilizer recommendation once you have the soil test values for wheat.Soil pH and EC both need to be analysed since they will give you fundamental reasonxs about your soil..
For analysis of the available macro and micro-nutrient that are related to crop growth and nutrient uptake, sampling is recommended to a depth of 30 cm i.e. 6 inches. So, your soil depth is acceptable. You can use that sample for estimation of pH. You may also estimate for EC. I do not have the knowledge of what cultivars of wheat are growing under Multan area but hardly you find any cultivar of wheat roots who crossed 30 cm depth, so probably you need not have to wary about the depth of the root, you may use whatever the varieties you like to compare.

I agree with Srivastava sir's comments. However you can go up to 12 inches. Since in general wheat root growth will be up to 30 cm, hence 12 inches would be ideal. As mentioned you can go as 0-15 cm as surface soil and 15 to 30 cm as sub surface soil. It will give you a clear picture about the difference in layers also
