With starting 2% initial concentration of dextrose in fermentation broth. How I maintain minimum dextrose concentration as per OD or biomass gm/l.
There is any thumb rule for that or I have to do some experiments for that. There is consumption of ammonia hydroxide in fermentation broth so feed rate of dextrose how effect the ammonia consumption rate.
Depending upon the fermenter volume or the duration of feeding,one can choose the glucose concentration as 10, 20 or 40 %, preferably autoclaved at 121 degree centigrades for 15 mins.. there is a possibility of charring of glucose if you exceed tempearure and time of autoclaving.or may be filter sterilized although 40 % may be too viscous for this purpose....I hope I made my answer clear

The dextrose rate or concentration is not everytime constant and differ to state of E. coli. So it will vary greatly over the course of a run depending upon the bacterial growth. Hexose is one of the primary molecules which serve as energy sources for almost all organisms, including bacteria.Glucose feeding can be set to respond to other sensor cues like D.O. level, the pH reading, the turbidity measurement etc.
To answer your question the proper concentration of hexose feed is less than 50% at 32°C to 35°C with pH 7 to 7.2.
