Your question is not well elaborated. But in general. Drug loading in nanoparticles depends on the following factors;

Which kind of nanoparticles have you made for loading the drug, e.g metallic NP's, inorganic NP's etc?

What is the morphology of your NP's shown in SEM/TEM, e.g hollow or mesoporous etc.

Particle Surface area of your NP's.

Your nanoparticles better encapsulate hydrophilic drugs or hydrophobic drugs?

Either you want to load drug during the synthesis of nanoparticles or after the synthesis? etc

Thanks alot for a detailed description about all the terminologies that i was confused with. After reading you reply, i know that i have few characterizations left out before proceeding further.  The surface area should larger for higher drug loading 

i wnat to load drug after and also want to check drug laoding percentage in uv but i am facing problem is that absorbance is going high in samle then standard why ?
