Hi Jyotsna Singh, I am familiar with the salting-out method for DNA isolation and have always used Triton X (1%) instead of NP-40. However, if you still want to go ahead with NP-40 then use around 2-5% of it.

Jyotsna Singh Both Triton X and NP40 are commonly used detergents. The percentages I have mentioned is what works well and is being used routinely in our lab. For your purpose I would suggest you to use 1% triton X because if you go any higher it would be harsh on your DNA.
