Extraction of polysaccharides from plant source?
Hi, I am trying to extract a polysaccharide from a plant source, I need to get it pure as much as possible, I do remove the fats by treating with hexane, and want to remove proteins and other materials with keeping the hydrogel properties, anybody can help in this?

You can remove proteins from the fat free plant material that is at your disposal by making it alkaline to a pH of about 9.00 -9.2 and carrying out filtration to remove solubilized proteins. Wash it (insoluble mass) thoroughly to make it free from any protein residue at the same pH and then wash it to make it free from alkali. The mass that you get is polysaccharides including starch, cellulose, hemicellulose along with lignin and the mineral matter associated with the bio-polymers.
To remove the mineral constituent you can think of giving ion-exchange treatment if it really works out in this case.

Thanks Arun, the problem is I can't do filtration because of the high viscosity of the "hydrogelly" material that results after treatment with aqueous solutions!

Dear Asim,
In that case don't use aqueous alkali solution and replace some water with organic solvent such as ethanol/methanol.
If you want to avoid use of alkali altogether then to get rid of proteins you can think of various proteolytic enzymes. Bacterial proteases with pH optima in the acid range or pepsin which is active at a pH as low as 2.0.
Prepare aqueous extract by cold maceration method and then remove tannin by 10% gelatine, filter and collect filtration then remove protein if it is present in cold macerate.
From the filtrate you can precipitate out the polysaccharide by addition of excess of acetone, thenfilter and collect the ppts
