has any body tried using Qaigen stool mini kit for extracting Cryptosporidium oocysts directly from feacal. IIf so, what is the concentration you usually getting since I have tried using it and it gave me very lower concentration (2-10ng)?

I have not tried using Qiagen kit in this way, however, you can use two-phase sucrose gradient concentration method in this regard.

Thanks Afshin . I am actually now strugling to know why when I measure the DNA concentration using nano drop, it gives me very low concentration although the purity was very good.

If the number of Cryptosporidium oocyst is very low, the low concentration of DNA is normal. I experienced this more and more. If you want this for PCR, you can do and may be in Primary PCR you didn't see any bands but in nested PCR you can see.
