Hi all, I'm trying to isolate exosomes using Amicon UFC filters. To do that, I'm using media collected from HUVECs. Has anybody tried this type of isolation or know how it works? Please let me know.
Shalini Paul, Amicon filters can be very useful for EV isolation (MWCO 30,000-100,000 Da typically). We use this step in our protocols very often. However, even for cell culture media, it may not be enough. You may want to check the purity of your EVs after ultrafiltration with fluorescent NTA and particle/protein ratio to see if you need to include any additional steps, like SEC or affinity purification. And don't forget to use serum-free media for EV collection.
Hi Daria, based on your experience, how many times is it possible to use Amicon® Ultra 4 ml with a limit of 100 kDa for EV concentration? What protocol do you use for cleaning of Amicon? Thank you in advance for your reply.
