I am extracting sulfated polysaccharide from various algae and I would like to add an alkali treatment to some my extraction protocols (notably to remove certain impurities).

I have found that strong alkali treatment could cause damages to polysaccharides, which range of concentration of NaOH will start be significant ? (I know it will depend of the polysaccharide properties, but I would like to have an idea of which range it does for sure, and which range in does not to stay safe). Is 0.1M good ? 1M ? Other parameter such as time of contact temperature also play a role here. Would 1h at 1M at RT be enough to induce a lot of damage ?

Finally, some of the polysaccharides I am extracting are sulfated, would alkali treatment affects the sulfate group on the polysaccharide ? I can not find proper literature on the topic.
