i came by a research paper the author of which attributed the death of a rat in their experimental group to the re-perfusion method . I myself have been having a very high rat of mortality in my experimental group after MCAO (1 hour ischemia). The mortility is so high thatthe animal did not even qualify the minimum 4 hour reperfusion period for TTC staining.The question is how to minimize the mortility and also that how is the method of reperfusion related to the mortility?
What anesthetic are you using? Do you have mortality in animals under similar conditions when you do a sham MCAO? Whose MCAO method are you using?
What paper did you read that had mortality after MCAO reperfusion?
dear sir Elwood we are using chloral hydrate 330mg/kg and we are following koizmi method. We have no mortality in the shame operated group. further i am using silicon coated nylone 4 suture of doccol. i use the tip diameter of 0.39mm for a 300gm rate while a tip diameter of 0.41mm for 330g and above.
Have you examined the entry site on the carotid artery to be certain there is not hemorage of blood after removal of the nylon monfilament? Do you have any rupture or bleeding at MCAO location when you examine the brain? Could you use gaseous anesthetic and improve the survival?

dear sir Dr. walls we were having hemorrhage initially, after which we modified our insertion technique so that our tip did not physically touch the origin of MCA,rather it was few milimeters from it. we used the animal weight to predict this distance. we then overcome hemorrhage but still had high mortality. it is worth noting that in the optimization phase of our animal model, we did not have sutures from doccol.we used heat blunted nylone 4 sutures and we had zero mortility at that time. i assumed that the size of infarct due to silicone coated suture is what causing all this.but since (in my experience) infarct can not be visualized inn the very early phase of reperfusion, so we could not rely on results from a model that did not survive longer,although animals which survived longer enough for the visualization of the infarct were having an infarct upto 40% of the contralateral hemisphere.
From my own experience, if animal weight is too small, mortality will increase. Also, if the operating temperature around the animal is not well-controlled or anaesthesic is not properly given, the mortality could increase. In addition, if your experiment includes additional pathology e.g. diabetes, mortality will be high.
