Ive been using sodium citrate and acd-a collection tubes to spin blood and extract platelets for PRP. The collection tubes I use do not have the separating gel. Ive noticed that in order to get all the platelets sitting right above the RBC I always end up extracting quite a bit of RBC. Im worried that the RBC will hinder the results after injection. I think PRP must be RBCs free to achieved better result. You improve PRP preparation method. You first cit-rated blood centrifuge at low RPM (1500 rpm) for 5 minutes. Transfer supernatent into other plane tube with out buffy coat. Again centrifuge at high RPM (4500) for 5 minutes. If total 5 mL than discord supernatent 4 mL you will yield 1 mL of PRP with out RBC contamination.
RBC in PRP is a contamination and it should be free of RBC.
When using commercial kits, this method is not convenient since every kit is expensive. What would you recomment when using commercials kits and you want to get rid of RBC on PRP?
