A research protocol on Ph.D degree titled: inhibition and degradation of aflatoxin production by probiotics.
This protocol would deals with the following:
1- Isolation of fast growing and acid producers probiotics such as lactobacilli,lactococci and pediococci onto MRS and M17 agar media, from fermented foods.
2-Preparation of aflatoxin producing fungal isolates.
3-Investigation the growth of aflatoxigenic fungi in presence of cell free supernatants from the above isolated probiotics( You should choose the strains with probiotic capability such as acid producers,bacteriocin producers,enzymes producers and the survived isolates with presence of bile acids and bile salts).
4-Compare between quantities of aflatoxin produced in control(no probiotic added)and in case of cell free supernatants added by different ratios).
5- Investigation of aflatoxin degradation by Gas chromatography .
6-In case of detecting aflatoxin degradation , take this original aflatoxin and add it in liquid medium and propagate this degrading probiotic onto this medium and calculate the aflatoxin subunits and their toxicity tests.
7- Search practically for the enzyme necessary for degradation of aflatoxin and produced by the probiotic.
