I work with murine tissue dissected from E11.5 mouse embryos. In the past, embryos were isolated from the uterus/ yolk sac in cold 1x PBS, washed and further dissected in cold 1x PBS. There was no problem with tissue survival in standard explant organ culture assays performed after dissection as described. In the last few months, more and more litters (from mice of different genetic backgrounds) have bad quality of embryonic organs already right after dissection (also confirmed by lysotracker assay) - for example in lungs, the surface of the organ looks rough and the tissue is more opaque than usual.

Anyone got ideas on what the problem might be or had a similar issue?
