Hello,
Currently I am trying with HeLa cells but in both cases I’m having problems with cell viability, so my cells are dying before I can see final results (seeding density for TCID50 96 wells plate: 2.5 x 10^4, and for pfu 24 wells plate: 2.5 x 10^5). I think I might use MRC5 cells for the next time (they live longer).

according my experience, you should use MEM 1X in PFU protocol, when the methylcellulose solidify add 100-500 uL of MEM (the volume of MEM depend of the plate size, is only to avoid drying). You can add drops of MEM each 24 or 48 h depending if the plate dry its
