I am studying the surface changes during bacterial degradation of rice straw using XPS. Do i need to do triplicates to assess the results? Is it the normal practice?
XPS is a surface analytical technique used for characterization of solid surfaces for identification and quantification of elements, and their oxidation state on the surface. What are you looking for surface degradation of rice straw by bacteria is very important. Please note that XPS is very sensitive to surfaces and characteristics of surface at different places of a sample will be different. If you are expecting change in elemental composition and oxidation state on the surface of rice straw by bacterial degradation, you can go for XPS, no need for triplicate. If you are looking for only surface degradation, then SEM-EDS is a suitable and faster technique which can provide morphological changes including elemental composition upon degradation by bacteria.
