Vanillin is soluble to about 10 g / L of water, so you'll need another solvent. It is soluble to ~50 mg/mL in ethanol. How sensitive is your assay to other solvents? I'd suggest DMSO but it seems vanillin isn't so soluble in this solvent.
I measured myself the antioxidant activity of vanillin using my own assay. The activity is very weak. Use lower concentrations of vanillin and check the activity. If it's zero, you can estimate the upper level of its activity.
@Jack silver, Thank you for your answer. But I can't use 50% ethanol because one of the solvent of my extract is 50% ethanol.So it will alter the actual result.
@Chaima Thanks. I will add 1500 microliter 37% HCl according to my protocol. Does it solve the issue? I initially tried to dissolve 4 g vanillin in 100 ml water but found it difficult to dissolve!
If you need to dissolve to an extent of 4% try in acidified condition with water . Make a pasty mass with HCl or H2SO4 then add water to hasten the dissolution
