Does tissue homogenization for hydroyproline colorimetric assay need protease inhibitor?

dear colleagues

i am using hydroxyproline colorimetric assay kit from biovision. can i homogenize my tissue samples in dH2O without protease inhibitor?

This is the protocol for the kit:

http://www.biovision.com/manuals/K555.pdf

It specifies homogenization in water. Protease inhibitor is unnecessary. The sample will be degraded into individual amino acids with concentrated HCl anyway.
