I am studying entrapment of a Cox-2 inhibitor in solid lipid nano-particles (SLNs). While making a standard curve, I used methanol-water mixture as a solvent (as the drug is sparingly water soluble). But my SLN suspension doesn't contain any methanol. Plus it contains traces of lipids and surfactants. Can I use the standard curve for the entrapment studies?
Yes you can the use the standard curve for your entrapment studies. At the moment you are not dealing with In vitro-In vivo correlation, so it is okay to use it.
You must determine if there is any absorption overlap between your methanol:water solvent alone and the COX inhibitor since methanol is not in the experimental group. I don't think methanol has any absorption peaks in the visible range, don't know about UV. I would honestly be more concerned about background absorption by your SLNs that are present in the experimental samples and not standard curve.
Thanks guys. Even I am of the same opinion. The absorption peak for methanol is below 203nm and that for the cox-2 inhibitor is around 252nm. So there is no problem of overlap. But i will have to check for SLNs.
