we are trying to prepare the Taq in our laboratory from cloned E. coli. To induce the enzyme we added IPTG in the Luria Broth (LB) after the growth of bacteria. The normal concentration of IPTG (0.1 M) used was 250 microlitre in 50 ml of LB. If we add increased concentration of IPTG (>250 microlitre), does it affect the growth of bacteria or Taq production?

Hi there,
What you call normal IPTG concentration is normally optimal IPTG concentration (resulting from the observation made on previous experiments). If it's actually optimal concentration then neither increase nor decrease in IPTG concentration would improve the level of expression of the Taq polymerase. On the contrary such variation may result in lower expression level.
