I am planning to run a glucose uptake experiment in mice (In vivo). I would like to inject H3-2DG and just measure the percent of H3 in different tissues. No clamp experiment, but I was thinkMing to inject H3-2DG (10microCurie per mice in a continuous infusion of 30min) with or without an insulin bolus at time 0 (0.5U/kg).
My questions are if this protocol makes sense or if you have any suggestion fir Improvement.

It depends what is your question ? when do you plan to sac the mice ? at the end of the infusion ?
When I did ITT (0.75U) in chow diet or high fat diet mice, the drop of glycemia was usually 30min after the bolus of insulin. 30 min of glucose infusion should be enough. However, the lonely experiment I have seen radioactive glucose infusion is during clamp experiment. I think that you can just try during a pilot experiment ?
