I work with obligate aerobic bacteria and need to check if they are motile. I was thinking of using SIM medium, but it occurred to me that it might not work with obligate aerobes. Can anyone clarify?
It can. However, instead of pouring it as a deep, pour it as a shallow plate (~1.5 to 2 mm deep). Then, inoculate into the center of the plate. You will be able to read all the characteristics except that you will use more Kovac's Reagent, obviously. However, if you are only interested in motility, a soft agar plate is the best way.
