I am working specifically on rice root proteins for proteomics. Now I am at the stage of standardization of extraction buffer for rice roots. I followed Raorane, Narciso, and Kohli 2016 (attached below) protocol. According to protocol heat treatment is included and I followed the same. I observed degradation of proteins (image attached). For reference I had even included rice shoot samples.

Can anyone please suggest me any specific working protocol for rice root total protein extraction?
