Does perfusd mouse brain need Heat Induced Epitope Retrieval (HIER) for IF with anti NeuN antibody?

Dear all,

I know for paraffin section it is necessary to perform Heat Induced Epitope Retrieval for IF. Does anyone have experience for the paraformaldehyde perfused mouse brain section? I am trying to stain the neurons with a anti NeuN antibody from Chemicon.

Yes; you need it.

I stained some sections in the past and I used Sodium citrate buffer for mouse brain tissue; you will have to check and optimize your times empirically.
