I used to work with a murine model of peripheral myelination that worked fine. Now i'm trying to set up a human primary schwann cell line (commercial, ScienceCell) that is able to differentiate and myelinate. I used murine NGF 2.5S for rat Schwann cell differentiation and worked fine, but now it does not seem to work in the human line. I've seen that both human and mouse NGF share 85% of the sequence, but maybe the cross-species differences make it less efficient for the human line? Has anybody worked with human schwann cell lines and used a specific protocol for them? Any thoughts?
