I want to mark the neurons I am recording from during whole cell Patch clamp. I am thinking of using Lucifer Yellow. Does it increase noise during recording? What are the better alternatives to Lucifer Yellow?
I used Lucifer Yellow in the intracellular solution during VC patch clamp recordings and didn't observe extra noise. But it is considered a ROS generator. Cosidering this, if I were you I would rather use something like Alexa 568 or any other fluorophores which do not interfere with your intrinsically expressed fluorescent proteins. Vendors like Thermo Fisher have several options that can potentially be fixed afterwards what makes them superior for post-hoc histology analysis.
We have used LY before but have gotten much better labeling of neurons using biocytin in the intracellular solution. It requires HIC staining but works pretty well.
