I have some stubs of plant tissues that are gold-coated. I was wondering if there is a possibility to do EDS analysis. Does the energy of the K or L edge of gold affect the analysis of the lighter elements? Thanks
Gold gives it's own peaks on spectrum and it will reduces intensity of peaks of other elements, especially light ones. But qualitative EDS is still possible, better for thin gold coating.

For the most sputter coater's settings time of 200 sec is very high. As Joao said, it is difficult to say about your settings, what time is the best. You can put a piece of writing paper in your coater and shield part of it with a nut or something (to have a contrast). The best time of coating - when unshielded part of paper became grayish, but not dark. For some specimens with highly developed topography this time may be not enough, but you always can add some more coating, and you cannot took away an excess of it.

Backscattered elétrons and Secondary electrons. What is the best form EDX and Why? The depth of Backscattered penetration is high? So I can get more signal What abou the noise?
It does not matter for EDS whether you use SE or BSE for observation. BSE could be handy if you have phases/ contaminations with different atomic numbers. Do not think it is applicable for plant tissues. By the way, why do you need EDS, what you hope to find? EDS is mostly noneffective for organic matter.
