I am working with differentiated PC12 cells and I have been asked to show that cells undergo apoptosis after 6 Hydroxy Dopamine treatment.
What is the best marker to show such event?
I have an antibody against PARP (113 kDa), but should I expect a decreasement of this protein in response to the treatment respect to control cells?

Normally, PARP maintains viability of cells, therefore decreasing in PARP expression may be reflect the cytotoxicity of 6-OHDA in the cells. But, you should use a antibody that targeting cleaved-PARP which is a marker of apoptotic activation. As you know, PARP is cleaved by active-caspases (e.g. CASP3) in response to toxic agents during apoptosis.

Treatment of PC12 cells with 6-OHDA induces apoptosis. The apoptotic phynotype is accompanied by caspase activation and PARP cleavage. So you do get a reduction in full length PARP and increase in cleaved form of pARP as a function of time.
